Ciprofloxacin induced antibiotic resistance in Salmonella Typhimurium mutants and genome analysis

Antibiotic resistance of Salmonella species is well reported. Ciprofloxacin the frontline antibiotic for salmonellosis. The repeated exposure to ciprofloxacin leads resistant strains. After 20 cycles exposure, bacterial clones were evaluated. colony size mutants was small and had an extended lag phase compared parent strain. whole genome sequencing showed 40,513 mutations across genome. Small percentage (5.2%) non-synonymous. Four-fold more transitions observed than transversions. Ratio < 1 transition vs transversion a positive selection trait. Mutation distribution uniform. native plasmid exception 2 on 90 kb plasmid. important genes like dnaE, gyrA, iroC, metH rpoB involved in point mutations. analysis revealed most metabolic pathways affected.

We characterized 44 Salmonella enterica serotype Typhimurium isolates from Tongji Hospital outpatients in Wuhan, China, May 2002-October 2005.
